Deiodinase, iodothyronine, type III encoded by this intronless gene (DIO3) belongs to the iodothyronine deiodinase family. It catalyzes the inactivation of thyroid hormone by inner ring deiodination of the prohormone thyroxine (T4) and the bioactive hormone 3,3',5-triiodothyronine (T3) to inactive metabolites, 3,3',5'-triiodothyronine (RT3) and 3,3'-diiodothyronine (T2), respectively. This enzyme is highly expressed in pregnant uterus, placenta, fetal and neonatal tissues, and thought to prevent premature exposure of developing fetal tissues to adult levels of thyroid hormones. It regulates circulating fetal thyroid hormone concentrations, and thus plays a critical role in mammalian development. Knockout mice lacking this gene exhibit abnormalities related to development and reproduction, and increased activity of this enzyme in infants with hemangiomas causes severe hypothyroidism. This protein is a selenoprotein, containing the rare selenocysteine (Sec) amino acid at its active site. Sec is encoded by the UGA codon, which normally signals translation termination. The 3' UTRs of selenoprotein mRNAs contain a conserved stem-loop structure, designated the Sec insertion sequence (SECIS) element, that is necessary for the recognition of UGA as a Sec codon rather than as a stop signal.
The DIO3 Polyclonal Antibody is a highly specific and sensitive tool designed for the detection and quantification of the DIO3 protein in various biological samples. This antibody has been meticulously developed using advanced techniques to ensure exceptional performance and reliability.
The DIO3 protein, also known as type 3 iodothyronine deiodinase, plays a crucial role in the regulation of thyroid hormone levels and metabolism. It is primarily expressed in the brain, placenta, and fetal tissues, and its dysregulation has been implicated in various pathological conditions, including neurodevelopmental disorders and cancer.
Our DIO3 Polyclonal Antibody exhibits exceptional specificity, binding exclusively to the DIO3 protein without cross-reactivity to other related proteins. This high specificity ensures accurate and reliable results, enabling researchers to confidently study the expression and function of DIO3 in various biological systems.
Furthermore, this antibody demonstrates remarkable sensitivity, allowing for the detection of low levels of DIO3 protein in even the most challenging samples. Its sensitivity is further enhanced by its ability to recognize both native and denatured forms of the DIO3 protein, making it suitable for a wide range of applications, including Western blotting, immunohistochemistry, and immunofluorescence.
The DIO3 Polyclonal Antibody is supplied as a ready-to-use solution, eliminating the need for time-consuming and error-prone antibody preparation. It is provided in a convenient format, ensuring ease of use and compatibility with various experimental protocols.
In summary, the DIO3 Polyclonal Antibody is a highly specific, sensitive, and reliable tool for the detection and quantification of the DIO3 protein. Its exceptional performance and ease of use make it an invaluable asset for researchers studying the role of DIO3 in various biological processes and diseases.
