Workflow (overview)

Prepare sample
Collect cell-culture supernatant and dilute with the supplied Sample Buffer to 10% (v/v).
Incubate (50°C)
Pre-warm a 0.2 mL heat block or thermal cycler to 50°C. Incubate the assembled test for exactly 10 minutes.
Flip & read
Flip per instructions, confirm fluid flow on the strip, then read results after ~2 minutes.

Sample types & prep tips

  • Cell culture supernatant: suitable for suspension and adherent cultures (test before passaging/media change).
  • FBS screening: check serum lots before use in critical workflows.
  • Suspension / semi-adherent: remove cells by centrifugation (500 g, 5 min) and test supernatant (per manufacturer guidance).
Highly confluent, long-maintained cultures may accumulate inhibitors that can affect assay performance (manufacturer note).
