Application: For quantitative detection of mouse TrkB in tissue lysates or cell culture supernates. The significance of detection in serum, body fluids is undetermined.

Introduction: TrkB receptor also known as TrkB tyrosine kinase or BDNF/NT-3 growth factors receptor or neurotrophic tyrosine kinase, receptor, type 2 is a protein that in human s is encoded by the NTRK2 gene. The TrkB receptor is part of the large family of receptor tyrosine kinases, it is located at the cellular membrane, and is activated by binding of a ligand to the receptor's extracellular domain. TrkB is the high affinity catalytic receptor for several "neurotrophins", which are small protein growth factors that induce the survival and differentiation of distinct cell populations.

Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-TrkB polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-TrkB polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the TrkB amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of TrkB can be calculated.
