Introduction: L-selectin, also known as CD62L, is a cell adhesion molecule found on lymphocytes. It belongs to the selectin family of proteins, which recognize sialylated carbohydrate groups. The molecule is composed of multiple domains: one homologous to lectins, one to epidermal growth factor, and two to the consensus repeat units found in C3/C4-binding proteins. L-selectin acts as a "homing receptor" for lymphocytes to enter secondary lymphoid tissues via high endothelial venules. Ligands present on endothelial cells will bind to lymphocytes expressing L-selectin, slowing lymphocyte trafficking through the blood, and facilitating entry into a secondary lymphoid organ at that point. It may also play a role in neutrophil adhesion to endothelium at sites of inflammation.

Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-L-Selectin polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-L-Selectin polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the L-Selectin amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of L-Selectin can be calculated.

Storage and Expiration: Store at 2-8℃ for 6 months, or at -20℃ for 12 months.
