Application: For quantitative detection of IL-13 in rat serum, plasma, body fluids, tissue lysates or cell culture supernatants.
Introduction: Interleukin 13 (IL-13) is a protein that in human s is encoded by the IL13 gene which maps to 5q23-q31. It is cytokine secreted by many cell types, but especially T helper type 2 (Th2) cells, that is a mediator of allergic inflammation and disease. IL-13 is associated primarily with the induction of airway disease, it also has anti-inflammatory properties. IL-13 induces many features of allergic lung disease, including airway hyperresponsiveness, goblet cell metaplasia and mucus hypersecretion, which all contribute to airway obstruction. It also induces secretion of chemokines that are required for recruitment of allergic effector cells to the lung.
Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-IL-13 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-IL-13 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washedaway with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the IL-13 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of IL-13 can be calculated.
Storage and Expiration: Store at 2-8℃ for 6 months, or at -20℃ for 12 months.
