Introduction: C-X-C motif chemokine 10 (CXCL10) also known as Interferon gamma-induced protein 10 (IP-10) or small-inducible cytokine B10 is an 8.7 kDa protein that in human s is encoded by the CXCL10 gene. The gene is located on human chromosome 4 in a cluster among several other CXC chemokines. CXCL 10 is a small cytokine belonging to the CXC chemokine family. It is secreted by several cell types in response to IFN-γ. CXCL10 has been attributed to several roles, such as chemoattraction for monocytes/ macrophages, T cells, NK cells, and dendritic cells, promotion of T cell adhesion to endothelial cells, antitumor activity, and inhibition of bone marrow colony formation and angiogenesis.

Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-CXCL10 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-CXCL10 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the CXCL10 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of CXCL10 can be calculated.

Storage and Expiration: Store at 2-8℃ for 6 months, or at -20℃ for 12 months.
