PEmax represents a significant architectural optimization of the PE2 system. By incorporating an additional Nuclear Localization Signal (NLS) and a refined nCas9 domain, it achieves superior nuclear import and targeting efficiency. Unlike traditional CRISPR methods, PEmax enables precise genomic modifications without inducing double-stranded breaks (DSBs), thereby maximizing editing yield and broadening the scope of targetable base conversions with enhanced fidelity. Synthesized via in vitro transcription with a Cap 1 structure and poly(A) tail, it mimics mature mRNA to enhance stability, translation efficiency, and performance while minimizing immune responses.
