What is single-cell sequencing?

Single-cell sequencing profiles DNA, RNA, chromatin, or other molecular features from individual cells. In widely used single-cell RNA-seq workflows, cell-specific barcodes allow pooled sequencing reads to be assigned back to their cell of origin—revealing distinct cell types, transient states, and rare populations within a heterogeneous sample.

Why counting and viability matter

A sequencing platform cannot correct a poorly characterized input. Cell concentration informs how much suspension to load; viability indicates how much of that input is biologically useful. Together, these measurements influence recovery, data quality, and efficient use of reagents.

  • Too concentrated: the probability of capturing more than one cell together can increase.
  • Too dilute: fewer cells may be recovered than planned.
  • Low viability: compromised cells can contribute ambient nucleic acids and background.
  • Inconsistent counts: sample comparisons and loading decisions become less dependable.
