Same Cytokine Concentration.
But Is the Biological Activity the Same?
Measuring cytokine concentration and measuring cytokine function answer two different questions. Reporter cell-based bioassays can evaluate whether a cytokine actually activates its receptor and downstream signaling pathway.
Concentration Is Not the Same as Biological Activity
Immunoassays such as ELISA are excellent tools for measuring the amount of cytokine present in a sample.
However, detection of a protein epitope does not necessarily demonstrate that the cytokine retains normal receptor binding and signaling activity.
Protein folding, aggregation, degradation and manufacturing differences can potentially influence biological potency even when measured protein concentrations appear similar.
How Does a Reporter Cell Bioassay Work?
A cytokine-responsive reporter cell line connects receptor activation to a measurable reporter gene.
Depending on the cytokine pathway, activation of NF-κB/AP-1, STAT3, STAT5 or another signaling pathway can drive expression of a reporter such as SEAP.
The resulting signal therefore reflects a functional sequence: receptor activation → intracellular signaling → reporter expression.
Example: Measuring IL-21 Bioactivity
InvivoGen HEK-Blue™ IL-21 cells are designed to monitor IL-21-induced STAT3 activation.
STAT3 activation drives production of a SEAP reporter, providing a convenient colorimetric readout of IL-21 stimulation.
A cytokine dose-response curve can therefore be used to compare bioactivity or relative potency between samples.
The Same Platform Can Evaluate Neutralizing Antibodies
Reporter assays can also be configured to measure inhibition.
When a neutralizing antibody blocks cytokine-receptor signaling, reporter activity decreases in a concentration-dependent manner.
This provides a functional assessment of whether an antibody actually inhibits cytokine signaling rather than simply binding the cytokine or its receptor.
