DNA methylation is an important form of epigenetic modification. In particular, covalent binding between cytosine's fifth carbon atom and the methyl group on genomic DNA is significant because it modifies cytosine to 5-methylcytosine (5mC). DNA methylation plays an important role in regulating gene expression, maintaining chromatin structure, gene imprinting, X chromosome inactivation, embryonic development, and other biological processes. It also has a profound impact on the occurrence/development of aging and diseases, with existing proof that DNA methylation is closely related to tumors, mental diseases, metabolic diseases, autoimmune diseases and many more. When bisulfite is used to treat DNA, unmethylated cytosine C goes through a series of sulfonation, deamination, desalting, and desulfonation reactions, ultimately converting unmethylated cytosine (C) to uracil (U), while methylated cytosine C remains unchanged during the conversion process.
