How to Select Capture and Detection Antibody Pairs for ELISA Development
Developing a reliable sandwich ELISA requires more than selecting two antibodies against the same target. The capture and detection antibodies must recognize compatible, non-overlapping epitopes.
Researchers developing a custom ELISA often need to identify an appropriate capture antibody and detection antibody pair.
Two antibodies against the same antigen are not automatically compatible. If both recognize the same or overlapping epitopes, simultaneous binding may be reduced or prevented.
How a Sandwich ELISA Works
Seven Factors to Review When Selecting an Antibody Pair
The capture and detection antibodies should recognize distinct, accessible epitopes.
Review whether each antibody is monoclonal, polyclonal, or recombinant.
Confirm compatibility with the species of your biological sample.
Consider serum, plasma, culture supernatant, lysate, or other intended matrices.
Check for BSA, glycerol, sodium azide, or other components that may affect downstream labeling.
Determine whether the detection antibody will be biotinylated, HRP-conjugated, or otherwise labeled.
Select a standard protein that contains the epitopes recognized by the antibody pair.
Monoclonal or Polyclonal Antibody Pair?
| Feature | Monoclonal-Based | Polyclonal-Inclusive |
|---|---|---|
| Specificity | High | Broader recognition |
| Epitope Recognition | Defined | Multiple possible |
| Reproducibility | High | Product dependent |
| Sensitivity | Pair dependent | Can be high |
When Do You Need a Carrier-Free Antibody?
BSA, glycerol, sodium azide, and other formulation components may interfere with certain antibody labeling or conjugation procedures.
For these workflows, consider whether the antibody can be supplied as carrier-free, BSA-free, or in a PBS-only formulation.
Can You Purchase an Antibody Pair Without a Complete ELISA Kit?
Yes, depending on the supplier. Researchers developing or optimizing their own assay may only require the capture antibody, detection antibody, and standard protein.
Some products can be supplied as individual reagents or as matched antibody pairs, providing more flexibility than a complete preconfigured kit.
