The composition of exosomes varies depending on their source, and their proteins are mainly divided into two major categories: membrane proteins and cytoplasmic proteins. According to literature statistics, CD9, CD63,CD81, and TSG101 are the four most frequently detected targets. Among them, CD63 is particularly important as it participates in various cellular processes and plays a key role in the protein sorting of late endosomes and multivesicular bodies, promoting the formation of exosomes. Other common targets include Alix, HSP70, Flotillin-1, and Syntenin-1, as well as CD82, HSP60, HSP90, LAMP2B, and LMP1, which are expressed in exosomes of different origins.
However, the markers of exosomes are not fixed and may require different combinations of markers depending on the cell type and the mechanism of exosome formation. Therefore, in specific studies, appropriate markers should be selected for analysis based on the cell type and conditions of exosome formation.
