Publication: Disulfide linkage-tethered zwitterion siRNA polyplexes targeting GalNT1/Tn antigen axis for colorectal cancer immunotherapy

This study introduced a disulfide-crosslinked zwitterionic siRNA polyplex targeting the GalNT1/Tn antigen axis in colorectal cancer. By combining GalNT1 silencing with serine depletion, the approach disrupted the immunosuppressive Tn–MGL axis, enhanced CD8+ T-cell infiltration, and inhibited tumor growth and metastasis.
