What Is an Antibody-Drug Conjugate?

An ADC is composed of three engineered components:

  • Antibody: Recognizes and binds a tumor-associated antigen on the cancer cell surface.
  • Linker: Connects the antibody and payload and controls payload release inside the target cell.
  • Cytotoxic payload: A highly potent drug that induces cancer cell death, often through DNA damage or microtubule disruption.

After the antibody binds to its target antigen, the ADC-antigen complex is internalized into the cancer cell. The linker then releases the cytotoxic payload, leading to apoptosis of the target cancer cell.
